Mouse GPNMB ELISA kit (ab270892) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of GPNMB protein in mouse serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate mouse GPNMB with 362 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
GPNMB (Glycoprotein non-metastatic gene B), also known as Osteoactivin, is a type I transmembrane glycoprotein most often associated with endosomal/lysosomal compartment and over expressed in many types of cancer. GPNMB is involved in many physiological processes, including transport of late melanosomes to keratinocytes, regulation of osteoblast and osteoclast, stimulation of dendritic cell maturation, and stimulation of adhesion of dendritic cells to endothelial cells.

The GPNMB standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of GPNMB were measured in duplicates, interpolated from the GPNMB standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 100% and plasma (EDTA) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean GPNMB concentration was determined to be 19.4 ng/mL in neat serum, 6.5 ng/mL in plasma (citrate) and 6.6 ng/mL in plasma (EDTA).
