Mouse Fas Ligand ELISA kit (ab270894) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fas Ligand protein in mouse serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate mouse Fas Ligand with 5.7 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Fas Ligand (FasL), also known as Tumor necrosis factor ligand superfamily member 6 or CD95 ligand is a cytokine that binds to TNFRSF6/FAS/CD95, a receptor that transduces the apoptotic signal into cells. Fas Ligand is involved in cytotoxic T-cell-mediated apoptosis, natural killer cell-mediated apoptosis and in T-cell development. It also initiates fratricidal/suicidal activation-induced cell death (AICD) in antigen-activated T-cells contributing to the termination of immune responses. TNFRSF6/FAS-mediated apoptosis has also a role in the induction of peripheral tolerance. Rat Fas Ligand has 93% amino acids sequence identity with mouse Fas Ligand.

Function
Cytokine that binds to TNFRSF6/FAS, a receptor that transduces the apoptotic signal into cells. May be involved in cytotoxic T-cell mediated apoptosis and in T-cell development. TNFRSF6/FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. Binding to the decoy receptor TNFRSF6B/DcR3 modulates its effects. -

The Fas Ligand standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Fas Ligand were measured in duplicates, interpolated from the Fas Ligand standard curves and corrected for sample dilution. Undiluted samples are as follows: treated and untreated spleen supernatants 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fas Ligand concentration was determined to be 38 pg/mL in treated spleen supernatant and undetectable in untreated spleen supernatant.
