Mouse ECM1 ELISA Kit (ab252368) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of ECM1 protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse ECM1 with 19 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
ECM1 is a glycoprotein involved in the modulation of bone mineralization and plays a role in wound healing, scarring, and dermal integrity. Mutations of the ECM1 gene is the basis for lipoid proteinosis, a rare dermatologic disorder. ECM1 is currently being investigated for its role in breast cancer metastasis.

Function
Involved in endochondral bone formation as negative regulator of bone mineralization. Stimulates the proliferation of endothelial cells and promotes angiogenesis. Inhibits MMP9 proteolytic activity. -
Tissue specificity
Expressed in breast cancer tissues. Little or no expression observed in normal breast tissues. Expressed in skin; wide expression is observed throughout the dermis with minimal expression in the epidermis. -
Involvement in disease
Defects in ECM1 are the cause of lipoid proteinosis (LiP) [MIM:247100]; also known as lipoid proteinosis of Urbach and Wiethe or hyalinosis cutis et mucosae. LiP is a rare autosomal recessive disorder characterized by generalized thickening of skin, mucosae and certain viscera. Classical features include beaded eyelid papules and laryngeal infiltration leading to hoarseness. Histologically, there is widespread deposition of hyaline material and disruption/reduplication of basement membrane. -

The concentrations of ECM1 were measured in duplicates, interpolated from the ECM1 standard curves and corrected for sample dilution. Undiluted samples are as follows: mouse serum 1:4, mouse plasma (heparin) 1:5, mouse plasma (citrate) 1:5.7, mouse plasma (EDTA) 1:5, and 20% mouse lung supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ECM1 concentration was determined to be 11.60 ng/mL in mouse serum, 16.53 ng/mL in mouse plasma (heparin), 19.12 ng/mL in mouse plasma (citrate) , 24.39 ng/mL in mouse plasma (EDTA), and 11.32 ng/mL in mouse lung supernatant.
