Mouse CD40L ELISA kit (ab275105) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse CD40L protein in serum, plasma, and cell culture samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse CD40L with 11.77 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
CD40L is a cytokine involved in immunoglobulin class switching, B-cell proliferation, and IgE production. CD40L is primarily expressed on CD4+ T cells, although it is present in a wide variety of other cell types, and is also secreted. CD40L is a potential biomarker for atherosclerotic instability.

Function
Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL-4. Involved in immunoglobulin class switching.
Release of soluble CD40L from platelets is partially regulated by GP IIb/IIIa, actin polymerization, and an matrix metalloproteinases (MMP) inhibitor-sensitive pathway. -
Tissue specificity
Specifically expressed on activated CD4+ T-lymphocytes. -
Involvement in disease
Defects in CD40LG are the cause of X-linked immunodeficiency with hyper-IgM type 1 (HIGM1) [MIM:308230]; also known as X-linked hyper IgM syndrome (XHIM). HIGM1 is an immunoglobulin isotype switch defect characterized by elevated concentrations of serum IgM and decreased amounts of all other isotypes. Affected males present at an early age (usually within the first year of life) recurrent bacterial and opportunistic infections, including Pneumocystis carinii pneumonia and intractable diarrhea due to cryptosporidium infection. Despite substitution treatment with intravenous immunoglobulin, the overall prognosis is rather poor, with a death rate of about 10% before adolescence. -

The concentrations of CD40L were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, plasma (heparin) 50%, and plasma (EDTA) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
