This kit utilizes the sensitive substrate GlucGreen™ to monitor beta-glucosidase activity in the lysosomes of live cells.
Lysosomal beta-glucosidase enzyme is currently indicated to be involved in diseases such as Gaucher and Parkinson’s diseases. LysoLive™ Lysosomal beta-Glucosidase Assay Kit utilizes a substrate which only becomes fluorescent upon cleavage by the enzyme meaning lysosomes with low beta-Glucosidase (GBA) activity will exhibit reduced staining in a manner proportional to the enzyme activity levels in the cell.

Components 1 units LysoLive™ GlucGreen 5 x 10µl Opti-Klear™ Live Cell Imaging Buffer 1X 2 x 30ml

5µM LysoLive™ GlucGreen™, 16 hour incubation.

Media was then replaced with Opti-Klear Live Cell Imaging buffer containing Hoechst 33342 at 10 µg/ml.

LysoLive GlucGreen was added at a final concentration of 5µM in complete media and incubated for 16 hours. Next day, cells were washed once with 1X PBS and bathed cells in Opti-Klear Imaging Buffer containing Hoechst 33342 healthy leukocytes (bottom) were treated with CBE for 16 hours in complete media containing 5uM final concentration LysoLive GlucGreen. Next day, cells were washed once with 1x PBS and bathed cells in Opti-Klear Imaging Buffer containing Hoechst 33342.
