Human YB1 ELISA Kit (YBX1) (ab269544) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of YB1 (YBX1) protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human YB1 (YBX1) with 0.85 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Y box binding protein 1 (YB1) or nuclease-sensitive element-binding protein 1 is a DNA- and RNA- binding protein encoded by the YBX1 gene. YB1 binds preferentially to the 5'-[CU]CUGCG-3' RNA motif and specifically recognizes mRNA transcripts modified by C5-methylcytosine (m5C). YB1 is involved in translational repression, RNA stabilization, mRNA splicing, DNA repair, and transcription regulation.

Mediates pre-mRNA alternative splicing regulation. Binds to splice sites in pre-mRNA and regulates splice site selection. Binds and stabilizes cytoplasmic mRNA. Contributes to the regulation of translation by modulating the interaction between the mRNA and eukaryotic initiation factors (By similarity). Regulates the transcription of numerous genes. Its transcriptional activity on the multidrug resistance gene MDR1 is enhanced in presence of the APEX1 acetylated form at 'Lys-6' and 'Lys-7'. Binds to promoters that contain a Y-box (5'-CTGATTGGCCAA-3'), such as MDR1 and HLA class II genes. Promotes separation of DNA strands that contain mismatches or are modified by cisplatin. Has endonucleolytic activity and can introduce nicks or breaks into double-stranded DNA (in vitro). May play a role in DNA repair. Component of the CRD-mediated complex that promotes MYC mRNA stability.
The secreted form acts as an extracellular mitogen and stimulates cell migration and proliferation. -

Predominantly cytoplasmic in proliferating cells. Cytotoxic stress and DNA damage enhance translocation to the nucleus. Localized with DDX1, MBNL1 and TIAL1 in stress granules upon stress. Secreted by mesangial and monocytic cells after inflammatory challenges. The YB1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

YB1 in human Hela cell extract based on a 25 μg/mL extract load. The concentrations of YB1 were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
