Product overview
Human VEGF-D ELISA Kit (ab233625) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of VEGF-D protein in hep plasma, serum, cell culture supernatant, and cit plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human VEGF-D with 75.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Function
Growth factor active in angiogenesis, lymphangiogenesis and endothelial cell growth, stimulating their proliferation and migration and also has effects on the permeability of blood vessels. May function in the formation of the venous and lymphatic vascular systems during embryogenesis, and also in the maintenance of differentiated lymphatic endothelium in adults. Binds and activates VEGFR-2 (KDR/FLK1) and VEGFR-3 (FLT4) receptors. -
Tissue specificity
Highly expressed in lung, heart, small intestine and fetal lung, and at lower levels in skeletal muscle, colon, and pancreas. -
Sequence similarities
Belongs to the PDGF/VEGF growth factor family. -
Post-translational
modificationsUndergoes a complex proteolytic maturation which generates a variety of processed secreted forms with increased activity toward VEGFR-3 and VEGFR-2. VEGF-D first form an antiparallel homodimer linked by disulfide bonds before secretion. The fully processed VEGF-D is composed mostly of two VEGF homology domains (VHDs) bound by non-covalent interactions. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of VEGF-D were measured in duplicates, interpolated from the VEGF-D standard curves and corrected for sample dilution. Undiluted samples are as follows: serum neat, plasma (citrate) neat, and plasma (heparin) neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean VEGF-D concentration was determined to be 2,105 pg/mL in neat serum, 1,349 pg/mL in neat plasma (citrate) and 1,232 pg/mL in neat plasma (heparin). Native signal is undetectable in RPMI cell culture supernatant containing 10% bovine serum (not shown).

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean VEGF-D concentration was determined to be 2,193 pg/mL with a range of 956.9 – 3,270 pg/mL.
