Human Transferrin Receptor ELISA kit (ab272469) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Transferrin Receptor protein in serum, plasma, urine, saliva, cell culture supernatant and cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Transferrin Receptor with 18 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Transferrin receptor, also known as CD71 TfR1, is a 760 amino acid, transmembrane glycoprotein of plasma membrane composed of two disulfide-linked monomers encoded by the gene TFRC. Transferrin receptor is necessary to regulate iron transport into cells from transferrin by receptor mediated endocytosis. The soluble form of transferrin receptor is due to the proteolysis of Arg-100 in the extracellular domain. Measurements of soluble transferrin receptor may be an indication of iron deficiency due to chronic diseases

Function
Cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied transferrin receptor into specialized endosomes. Endosomal acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. Transferrin receptor is necessary for development of erythrocytes and the nervous system (By similarity). A second ligand, the heditary hemochromatosis protein HFE, competes for binding with transferrin for an overlapping C-terminal binding site. Positively regulates T and B cell proliferation through iron uptake (PubMed:26642240).

The concentrations of Transferrin Receptor were measured in duplicates, interpolated from the Transferrin Receptor standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:1,000, plasma (citrate) 1:1,000, plasma (EDTA) 1:500 and plasma (heparin) 1:1,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Transferrin Receptor concentration was determined to be 2,122 ng/mL in neat serum, 2,234 ng/mL in neat plasma (citrate), 1,714 ng/mL in neat plasma (EDTA) and 1,990 ng/mL in neat plasma (heparin).
