Human TFPI2 ELISA kit (ab275908) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human TFPI2 protein in human serum, plasma-citrate, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human TFPI2 with 16 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
TFPI2, Tissue factor pathway inhibitor 2 also known as placental protein 5 (PP5), is a serine protease inhibitor that plays a key role in plasmin-mediated remodeling of the matrix by inhibiting trypsin and plasmin. TFPI2 is involved in matrix homeostasis, fetal development, and preventing tumor metastasis through invasion. Hypermethylation of this protein is associated with many cancer types, including, colorectal, pancreatic, and cervical cancers. The immunogen used to create the antibodies for this kit represents the mature form of the protein. The protein homologs in mouse in rat share little sequence homology, and as such, are not expected to cross-react with this kit.

The TFPI2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of TFPI2 were measured in duplicates, interpolated from the TFPI2 standard curves and corrected for sample dilution. Undiluted samples are as follows: pregnant serum 1:100, pregnant plasma (citrate) 1:100, and HeLa cell culture supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TFPI2 concentration was determined to be 294.3 ng/mL in pregnant serum, 347 ng/mL in pregnant plasma (citrate), and 1.6 ng/mL in HeLa cell culture supernatant.
