Human RAB8 ELISA Kit (ab255718) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of RAB8 protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human RAB8 with 0.024 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering, and fusion. Rab proteins are highly conserved across species; human RAB8, also known as Ras-related protein Rab-8A, is 97.5% identical on amino acids level to mouse RAB8. Mouse and rat RAB8 proteins are 100% identical.

May be involved in vesicular trafficking and neurotransmitter release. Together with RAB11A, RAB3IP, the exocyst complex, PARD3, PRKCI, ANXA2, CDC42 and DNMBP promotes transcytosis of PODXL to the apical membrane initiation sites (AMIS), apical surface formation and lumenogenesis. Together with MYO5B and RAB11A participates in epithelial cell polarization. -

The RAB8 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of RAB8 were measured in duplicate and interpolated from the RAB8 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean RAB8 concentration was determined to be 2.16 ng/mL in HeLa cell extract, 1.16 ng/mL HepG2 cell extract, and 4.82 ng/mL in human colon tissue extract.
