Human PAI1+tPA ELISA Kit (ab192151) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PAI1+tPA protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human PAI1+tPA with 16 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Tissue plasminogen activator (tPA or PLAT) is a serine protease that catalyzes the conversion of plasminogen to plasmin, which is integral to blood clot breakdown. Plasminogen activator inhibitor-1 (PAI-1 or Serpin E1) is a serine protease inhibitor that blocks the function of tPA. PAI-1 and tPA form a complex that is stable and non-covalent. The PAI-1/tPA complex appears to be predictive of risk for myocardial infarction. This assay uses a PAI-1 specific capture antibody and a tPA-specific detector antibody to specifically detect PAI1/tPA complexes in Human biological samples. The reference protein is a non-covalent complex of PAI 1 and tPA.

The concentrations of PAI1-TPA complex were measured in duplicate and interpolated from the PAI1-TPA complex standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PAI1-TPA complex concentration was determined to be 2,040 pg/mL in serum and 2,160 pg/mL in plasma (heparin).

Serum from 10 apparently healthy male donors was measured in duplicate. The mean PAI1-TPA complex concentration was determined to be 4,126 pg/mL with a range of 2,764- 5,584 pg/mL in male donors.

Recombinant proteins for PAI1, TPA, and PAI1-TPA complex were tested in PAI1 SimpleStep ELISA, TPA SimpleStep ELISA and PAI1-Complex SimpleStep ELISA to determine specificity of the PAI-TPA complex ELISA. Background-subtracted data values are graphed. The PAI1-TPA Complex assay does not detect free PAI1 or TPA protein.

PAI1-TPA complex, PAI1 protein and TPA protein were measured in 8-fold diluted serum and plasma. PAI1, TPA, and PAI1-TPA complex values were measured and interpolated from their respective Standard Curve diluted in Sample Diluent NS or Sample Diluent 25BP. Mean +/-SD, n=2, are graphed.
