Human N-Cadherin ELISA Kit (ab254512) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of N-Cadherin protein in edta plasma, hep plasma, cell culture extracts, serum, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human N-Cadherin with 41 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
N-Cadherin is a member of the calcium-dependent adhesion molecule family of classical cadherins encoded by the CDH2 gene. This transmembrane, homophilic glycoprotein often can be found concentrated at adherens junctions in most cell types. The function of N-Cadherin depends much on the cellular contents such as cancer metastasis, interactions with catenins in neutral tissues, and maintaining the structural integrity of the heart by mediating strong adhesion between cardiomyocytes. This kit is raised as an extracellular fragment of N-Cadherin.

Function
Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. CDH2 may be involved in neuronal recognition mechanism. In hippocampal neurons, may regulate dendritic spine density. -

The N-Cadherin standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of N-Cadherin were measured in duplicates, interpolated from the N-Cadherin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (EDTA) 50%, plasma (heparin) 50% and HeLa cell culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean N-Cadherin concentration was determined to be 17,850 pg/mL in serum, 19,670 pg/mL in plasma (EDTA) and 12,801 pg/mL in plasma (heparin) and 8391 pg/mL in HeLa cell culture supernatant.

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean N-Cadherin concentration was determined to be 13,895 pg/mL in HeLa cell extract.
