Product overview
Human Myosin light chain 3 ELISA Kit (ab246538) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Myosin light chain 3 protein in tissue extracts and cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Myosin light chain 3 with 89 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Involvement in disease
Defects in MYL3 are the cause of cardiomyopathy familial hypertrophic type 8 (CMH8) [MIM:608751]. Familial hypertrophic cardiomyopathy is a hereditary heart disorder characterized by ventricular hypertrophy, which is usually asymmetric and often involves the interventricular septum. The symptoms include dyspnea, syncope, collapse, palpitations, and chest pain. They can be readily provoked by exercise. The disorder has inter- and intrafamilial variability ranging from benign to malignant forms with high risk of cardiac failure and sudden cardiac death. CMH8 inheritance can be autosomal dominant or recessive.
Defects in MYL3 are the cause of cardiomyopathy familial hypertrophic with mid-left ventricular chamber type 1 (MVC1) [MIM:608751]. MVC1 is a very rare variant of familial hypertrophic cardiomyopathy, characterized by mid-left ventricular chamber thickening. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The Myosin Light Chain 3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of Myosin Light Chain 3 were measured in duplicate and interpolated from the Myosin Light Chain 3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Myosin Light Chain 3 concentration was determined to be 1538 pg/mL in heart extract and 2893 pg/mL in skeletal muscle extract.
