Human MDH1 ELISA Kit (cytosolic malate dehydrogenase) (ab255724) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MDH1 (cytosolic malate dehydrogenase) protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human MDH1 (cytosolic malate dehydrogenase) with 64 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
MDH1 is the cytosolic form of malate dehydrogenase, an enzyme that catalyzes the oxidation of malate to oxaloacetate while concomitantly reducing NAD+ to NADH. Its classical role is the malate:aspartate shuttle, where it facilitates the transfer of electrons produced during glycolysis into the mitochondria to produce ATP. Beyond its classical role, this enzyme may play a primary regulatory role in adipogenesis.

The MDH1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of MDH1 were measured in duplicates, interpolated from the MDH1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (citrate) 25%, plasma (heparin) 12.5% and plasma (EDTA) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MDH1 concentration was determined to be 15.35 ng/mL in serum, 21.69 ng/mL in plasma (citrate), 15.07 ng/mL in plasma (heparin) and 13.22 ng/mL in plasma (EDTA).
