Human MCP2 ELISA Kit (ab223856) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MCP2 protein in cell culture extracts, cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human MCP2 with 1.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Monocyte chemoattractant protein 2 (MCP2), known also as C-C motif chemokine 8 (CCL8) is chemotactic factor that attracts monocytes, lymphocytes, basophils and eosinophils. Chemokines and their receptors, in conjunction with adhesion molecules and their ligands, provide cues to enable tissue-specific homing of T cells in the steady state and during inflammation. The processed form MCP2(6-76) does not show monocyte chemotactic activity, but inhibits the chemotactic effect most predominantly of CCL7, and also of CCL2 and CCL5 and MCP2. MCP2 protein can bind heparin.

Function

The concentrations of MCP2 were measured in duplicates, interpolated from the MCP2 standard curves. Undiluted samples are as follows: serum 100%, plasma (citrate) 100%, plasma (heparin) 100% and plasma (EDTA) 100%. The interpolated values are plotted (mean +/- SD, n=2). The mean MCP2 concentration was determined to be 19 pg/mL in neat serum, 16 pg/mL in neat plasma (citrate), 18 pg/ml in neat plasma (heparin), and 15 pg/mL in neat plasma (EDTA).

Undiluted samples are as follows: PBMC treated supernatant 0.08%, PBMC untreated supernatant 0.08%. The mean MCP2 concentration was determined to be 80,341 pg/mL in neat treated PBMC supernatant and undetectable in untreated supernatants (not shown).
