Product overview
Human IFN alpha/beta R2 ELISA Kit (IFNAR2) (ab264610) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IFN alpha/beta R2 (IFNAR2) protein in hep plasma, serum, and cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IFN alpha/beta R2 (IFNAR2) with 3.72 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Human IFN alpha/beta R2 (IFNAR2) is a100 kDa glycoprotein. It associates with IFNAR1 to form the type I interferon receptor, a receptor for interferons alpha and beta. IFNAR2 is also involved in IFN-mediated STAT1, STAT2 and STAT3 activation. Isoform 1 and isoform 2 are directly involved in signal transduction due to their association with the TYR kinase, JAK1 and Isoform 3 is a potent inhibitor of type I IFN receptor activity. Human IFNAR2 shares 47%, 47%, and 57% sequence homology with mouse, rat, and cow, respectively. The antibodies in this product are raised to the extracellular region of IFN alpha/beta R2.

Function
Associates with IFNAR1 to form the type I interferon receptor. Receptor for interferons alpha and beta. Involved in IFN-mediated STAT1, STAT2 and STAT3 activation. Isoform 1 and isoform 2 are directly involved in signal transduction due to their association with the TYR kinase, JAK1. Isoform 3 is a potent inhibitor of type I IFN receptor activity. -
Tissue specificity
Isoform 3 is detected in the urine (at protein level). Expressed in blood cells. Expressed in lymphoblastoid and fibrosarcoma cell lines. -
Sequence similarities
Belongs to the type II cytokine receptor family. -
Post-translational
modificationsPhosphorylated on tyrosine residues upon interferon binding. Phosphorylation at Tyr-337 or Tyr-512 are sufficient to mediate interferon dependent activation of STAT1, STAT2 and STAT3 leading to antiproliferative effects on many different cell types.

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The IFN alpha/beta R2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
Interpolated concentrations of native IFN alpha/beta R2 in human serum and plasma - heparin samples.
The concentrations of IFN alpha/beta R2 were measured in duplicates, interpolated from the IFN alpha/beta R2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50% and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IFN alpha/beta R2 concentration was determined to be 5.51 ng/mL in serum and 4.96 ng/mL in plasma (heparin).

Interpolated concentrations of native IFN alpha/beta R2 in human THP-1 cell extract based on a 250 µg/mL extract load.
The concentrations of IFN alpha/beta R2 were measured in duplicate and interpolated from the IFN alpha/beta R2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IFN alpha/beta R2 concentration was determined to be 489.21 pg/mL in THP-1 cell extract.
