Product overview
Human Hemopexin ELISA Kit (ab221838) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Hemopexin protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Hemopexin with 370 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Hemopexin is a heme-binding plasma glycoprotein which, after haptoglobin, forms the second line of defense against hemoglobin-mediated oxidative damage during intravascular hemolysis. A decrease in plasma hemopexin concentration reflects a recent release of heme compounds in the extracellular compartment. Heme–Hemopexin complexes are delivered to hepatocytes by receptor-mediated endocytosis after which Hemopexin is recycled to the circulation. Studies indicated that increased Hemopexin level associate with minimal change disease (MCD), sporadic Alzheimer's disease (AD), heavy-drinking chronic alcoholics, hemolytic anemias, chronic neuromuscular diseases and acute intermittent porphyria.

Research areas
-
Function
Binds heme and transports it to the liver for breakdown and iron recovery, after which the free hemopexin returns to the circulation. -
Tissue specificity
Expressed by the liver and secreted in plasma. -

Belongs to the hemopexin family.
Contains 5 hemopexin-like domains. -
Post-translational
modificationsN- and O-glycosylated. O-glycosylated with core 1 or possibly core 8 glycans. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of hemopexin were measured in duplicates, interpolated from the hemopexin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:40,000, plasma (citrate) 1:20,000, plasma (EDTA) 1:20,000, and plasma (heparin) 1:20,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean hemopexin concentration was determined to be 1004 µg/mL in serum, 768 µg/mL in plasma (citrate), 875 µg/mL in plasma (EDTA) and 812 µg/mL in plasma (heparin).
-
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean hemopexin concentration was determined to be 1043 µg/mL with a range of 584 – 1316 µg/mL.
-
The concentrations of hemopexin were measured in duplicates, interpolated from the hemopexin standard curves and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean hemopexin concentration was determined to be 24.3 ng/mL in HepG2 supernatant.
