Human Folate Binding Protein (FBP) ELISA kit (ab277710) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Folate Binding Protein (FBP) protein in human serum, plasma, cell culture supernatant, urine, salive, and milk samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Folate Binding Protein (FBP) with 3.08 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Folate Binding Protein is a receptor involved in update of folic acid and reduced folates. FBP is primarily expressed on epithelial cells and is essential during early embryogenesis. Upregulation of FBP is seen in some cancers and can be a marker for ovarian cancer.

Function
Binds to folate and reduced folic acid derivatives and mediates delivery of 5-methyltetrahydrofolate to the interior of cells. -
Tissue specificity
Exclusively expressed in tissues of epithelial origin. Expression is increased in malignant tissues. Expressed in kidney, lung and cerebellum. -
Involvement in disease
Defects in FOLR1 are the cause of neurodegeneration due to cerebral folate transport deficiency (NCFTD) [MIM:613068]. NCFTD is an autosomal recessive disorder resulting from brain-specific folate deficiency early in life. Onset is apparent in late infancy with severe developmental regression, movement disturbances, epilepsy, and leukodystrophy. Note=Recognition and diagnosis of this disorder is critical because folinic acid therapy can reverse the clinical symptoms and improve brain abnormalities and function. -

The concentrations of Folate Binding Protein were measured in duplicates, interpolated from the Folate Binding Protein standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100%, plasma (citrate) 100%, plasma (heparin) 100% and plasma (EDTA) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Folate Binding Protein concentration was determined to be 504.27 pg/mL in serum, 294.18 pg/mL in plasma (citrate), 355.36 pg/mL in plasma (heparin), and 290.02 pg/mL in plasma (EDTA).

Undiluted samples are as follows: HeLa supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Folate Binding Protein concentration was determined to be 151.87 pg/mL in HeLa supernatant.

Undiluted samples are as follows: urine 1: 50, saliva 1: 50, and milk 1: 800. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Folate Binding Protein concentration was determined to be 12.17 ng/ml in urine, 20.41 ng/ml in saliva, and 499.18 ng/ml in milk.
