Human ErbB 3 ELISA kit (ab275101) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human ErbB 3 protein in serum, plasma, cell culture supernatant, urine, saliva and cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ErbB 3 with 27.6 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Receptor tyrosine-protein kinase ErbB 3, also known human epidermal growth factor receptor 3 (HER3), is encoded by the ERBB3 gene in which is a member of the EGFR family. ErbB 3 is a type I transmembrane glycoprotein that binds to neuregulins NRG1 and NRG2, but lacks a working kinase domain. ErbB 3 is capable of heterodimerization of its fellow ErbB family members, most notably with ErbB 2 for signaling. Moreover, ErbB 3 is vital in the development of Schwann cells, neural crest differentiation, sympathetic nervous system, and heart values. Multiple studies have shown ErbB 3 to play roles in cellular growth, proliferation, differentiation, promotion and metastasis of cancer, and chemotherapeutic resistance.

Defects in ERBB3 are the cause of lethal congenital contracture syndrome type 2 (LCCS2) [MIM:607598]; also called Israeli Bedouin multiple contracture syndrome type A. LCCS2 is an autosomal recessive neurogenic form of a neonatally lethal arthrogryposis that is associated with atrophy of the anterior horn of the spinal cord. The LCCS2 syndrome is characterized by multiple joint contractures, anterior horn atrophy in the spinal cord, and a unique feature of a markedly distended urinary bladder. The phenotype suggests a spinal cord neuropathic etiology. -

The ErbB 3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of ErbB 3 were measured in duplicates, interpolated from the ErbB 3 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, plasma (heparin) 25% and MCF-7 supernatant neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ErbB 3 concentration was determined to be 4,877 pg/mL in serum, 3,874 pg/mL in plasma (citrate), 4,148 pg/mL in plasma (EDTA), 3,783 pg/mL in plasma (heparin), and 301.2 pg/mL in MCF-7 supernatant.
