Human eNOS ELISA Kit (ab263878) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of eNOS protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human eNOS with 77 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
eNOS, or NOS3, is an endothelial enzyme which produces nitric oxide and is the constitutively activated form found primarily in the vasculature. The nitric oxide produced by this enzyme regulates vascular tone and platelet aggregation. The standard protein in this kit is full length eNOS.

Function
Produces nitric oxide (NO) which is implicated in vascular smooth muscle relaxation through a cGMP-mediated signal transduction pathway. NO mediates vascular endothelial growth factor (VEGF)-induced angiogenesis in coronary vessels and promotes blood clotting through the activation of platelets.
Isoform eNOS13C: Lacks eNOS activity, dominant-negative form that may down-regulate eNOS activity by forming heterodimers with isoform 1. -

Variation in NOS3 seem to be associated with susceptibility to coronary spasm. -

Post-translational
modificationsPhosphorylation by AMPK at Ser-1177 in the presence of Ca(2+)-calmodulin (CaM) activates activity. In absence of Ca(2+)-calmodulin, AMPK also phosphorylates Thr-495, resulting in inhibition of activity (By similarity). Phosphorylation of Ser-114 by CDK5 reduces activity. -

Interpolated concentrations of native eNOS in human bEnd.3 cell extract based on a 40 µg/mL extract load.The concentrations of eNOS were measured in duplicate and interpolated from the eNOS standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean eNOS concentration was determined to be 5611.62 pg/mL.
