Human DKK3 ELISA Kit (ab236710) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of DKK3 protein in cit plasma, edta plasma, hep plasma, serum, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human DKK3 with 34.1 pg/ml sensitivity.

Function
Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease. -

The DKK3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of DKK3 were measured in duplicates, interpolated from the DKK3 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 20%, plasma (citrate) 10%, plasma (heparin) 10%, and plasma (EDTA) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DKK3 concentration was determined to be 38.4 ng/mL in serum, 30.5 ng/mL in plasma (citrate), 29.6 ng/ml in plasma (heparin), and 29.0 ng/mL in plasma (EDTA).

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DKK3 concentration was determined to be 19.8 ng/mL in HUVEC supernatant.
