Human Cytokeratin 8 ELISA Kit (ab267810) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Cytokeratin 8 protein in cell culture media, tissue extracts, and cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Cytokeratin 8 with 1.01 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Keratin, type II cytoskeletal 8 is a major component of the intermediate filament cytoskeleton and is a marker for epithelial cells. Abnormal expression of Cytokeratin 8 is found in various forms of cancer, and Cytokeratin 8 has been used as a marker for circulating tumor cells.

The Cytokeratin 8 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Interpolated concentrations of native Cytokeratin 8 in human MCF7, HepG2, and liver homogenate extract samples based on 6.25 µg/mL, 31.25 µg/mL, and 100 µg/mL extract loads, respectively.
The concentrations of Cytokeratin 8 were measured in duplicate and interpolated from the Cytokeratin 8 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Cytokeratin 8 concentration was determined to be 26.10 ng/mL in MCF7 extract, 10.03 ng/ml in HepG2 extract, and 6.71 ng/ml in liver homogenate extract.
