Human Cytokeratin 19 ELISA Kit (ab252367) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Cytokeratin 19 protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Cytokeratin 19 with 375 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Cytokeratin 19, also known as KRT19, is an intermediate filament protein that gives structure to epithelial cells. Beyond its role as a structural protein, KRT19 is involved in cell signaling pathways including Wnt/Notch and EGR1/PTEN/AKT, although the response to altered expression is highly cell-type dependent. Expression levels of this protein are altered in many types of cancers, and it is currently used as a biomarker for breast cancer.

Function
Involved in the organization of myofibers. Together with KRT8, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle. -
Tissue specificity
Expressed in a defined zone of basal keratinocytes in the deep outer root sheath of hair follicles. Also observed in sweat gland and mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, ectocervical epithelium (at protein level). Expressed in epidermal basal cells, in nipple epidermis and a defined region of the hair follicle. Also seen in a subset of vascular wall cells in both the veins and artery of human umbilical cord, and in umbilical cord vascular smooth muscle. Observed in muscle fibers accumulating in the costameres of myoplasm at the sarcolemma in structures that contain dystrophin and spectrin. -

The Cytokeratin 19 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Cytokeratin 19 were measured in duplicate and interpolated from the Cytokeratin 19 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Cytokeratin 19 concentration was determined to be 34.6 ng/mL .
