Human Chromogranin A ELISA kit (ab270885) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Chromogranin A protein in human serum, plasmas and cell culture supernatants. It uses our proprietary SimpleStep ELISA® technology. Quantitate human Chromogranin A with 20 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Chromogranin A (CHGA, CgA, also known as Parathyroid secretory protein 1) is a member of the granin family, which are acidic proteins found in the secretory granules of endocrine cells. CHGA consists of ~460 amino acids and is processed into numerous peptides that have diverse activities. Some of the peptides affect the secretion of other hormones, regulate metabolism, and regulate neural pathways that control behavior. CHGA has been shown to be elevated in the blood of cancer patients, to inhibit angiogenesis, and affect tumor response to chemotherapy. It is thought to be a biomarker for neuroendocrine tumors, and increased levels have been detected in patients with lung cancer, prostate cancer, and breast cancer. Elevated CHGA serum levels have also been detected in non-cancer disease state patients with heart and renal failure, hypertension, rheumatoid arthritis, and various other inflammatory diseases.

The Chromogranin A standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Chromogranin A (CHGA) were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (EDTA) 5%, plasma(citrate) 5%, plasma (heparin) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 56.7 ng/mL in serum, 48.5 ng/mL in plasma (EDTA), 53.0 ng/mL in and plasma (citrate), and 40.1 ng/mL in plasma (heparin).

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Chromogranin A (CHGA) concentration was determined to be 58.8 ng/mL with a range of 10.4 – 171.9 ng/mL.
