Human CD8A ELISA Kit (ab256393) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CD8A protein in cell culture supernatant, edta plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CD8A with 4.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Human CD8 alpha, or T-Cell surface glycoprotein CD8 alpha chain, is a cell surface glycoprotein encoded by the gene Cd8a. CD8A is found on cytotoxic T lymphocytes and helps mediates cell-cell interactions within the immune system. CD8 acts a coreceptor for MHC class 1/peptide complex either as a homodimer of two alpha chains or as a heterodimer of one alpha chain and one beta chain. CD8A interacts with LCK in a zinc-dependent manner, which initiates intracellular signaling pathways resulting in activation of cytotoxic T-lymphocytes.

Function
Identifies cytotoxic/suppressor T-cells that interact with MHC class I bearing targets. CD8 is thought to play a role in the process of T-cell mediated killing. CD8 alpha chains binds to class I MHC molecules alpha-3 domains. -
Involvement in disease
Defects in CD8A are a cause of familial CD8 deficiency (CD8 deficiency) [MIM:608957]. Familial CD8 deficiency is a novel autosomal recessive immunologic defect characterized by absence of CD8+ cells, leading to recurrent bacterial infections. -

The CD8A standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of CD8A were measured in duplicate and interpolated from the CD8A standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
