Human CA125 ELISA kit (ab274402) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human CA125 protein in serum, plasma, urine, cell culture supernatant, and milk. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CA125 with 0.02 U/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Cancer antigen 125 (CA125, also known as Mucin 16) is a membrane associated cell surface protein containing approximately 22,096 amino acids. CA125 is the largest member of the Mucin family of proteins and is highly O-glycosylated. The hydrophilic nature of mucin proteins gives them the ability to form protective/lubricating gel-like barriers to protect against chemotherapy agents, foreign particles and infectious agents. CA125 is used as a biomarker for ovarian cancer detection, with about 90% of women with advanced ovarian cancer having elevated levels of CA125 present in their blood serum.

The CA125 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of CA125 were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows: Hela cell culture supernatant, 50%, milk, 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 3 U/mL in Hela cell culture supernatant, 4.5 U/mL in milk.
