Human alpha-2 antiplasmin ELISA Kit (ab254502) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of alpha-2 antiplasmin protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human alpha-2 antiplasmin with 29.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Alpha-2-antiplasmin is a serine protease inhibitor. The mature form of alpha-2-antiplasmin is generated by cleaved at Pro-39 by both the prolyl endopeptidase FAP form and antiplasmin-cleaving enzyme FAP soluble form. The major targets of this inhibitor are plasmin and trypsin, but it also inactivates matriptase-3/TMPRSS7 and chymotrypsin.

Neurotrophic protein; induces extensive neuronal differentiation in retinoblastoma cells. Potent inhibitor of angiogenesis. As it does not undergo the S (stressed) to R (relaxed) conformational transition characteristic of active serpins, it exhibits no serine protease inhibitory activity. -

The alpha-2 antiplasmin standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of alpha-2 antiplasmin were measured in duplicates, interpolated from the alpha-2 antiplasmin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:20,000, plasma (citrate) 1:20,000, plasma (EDTA) 1:20,000, and plasma (heparin) 1:20,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha-2 antiplasmin concentration was determined to be 40.2 µg/mL in neat serum, 70.3 µg/mL in neat plasma (citrate), 59.4 µg/mL in neat plasma (EDTA), and 43.5 µg/mL in neat plasma (heparin).
