Arf6 Activation Assay Kit (ab211171) uses GGA3 PBD Agarose beads to selectively isolate and pull-down the active form of Arf from purified samples or endogenous lysates. Subsequently, the precipitated GTP-Arf is detected by western blot analysis using an anti-Arf6 antibody, which detects the human, rat, cow and dog protein.
Features: 1) non radioactive assay format; 2) fast results: 1 hour assay plus electrophoresis/blotting time; 3) includes Cdc42 positive control; 4) pink colored agarose beads for easy identification during washing and aspiration steps.

Small GTP-binding proteins (or GTPases) are a family of proteins that serve as molecular regulators in signaling transduction pathways.
Arf6 (ADP-ribosylation factor 6) is a 20 kDa protein, member of ARF gene family, which constitutes part of the Ras superfamily. The Arf proteis are categorized as class I (Arf1, Arf2, Arf3), class II (Arf4 and Arf5) and class III (Arf6), and they all share a common gene organization. Arf6 regulates a variety of biological response pathways that include protein trafficking, organelle structure, and cytoskeleton remodeling. Like other small GTPases, Arf6 cycles between an inactive GDP-bound form and an active GTP-bound form: in its active (GTP-bound) state, Arf1 binds specifically to the protein-binding domain (PBD) of GGA3 to control downstream signaling cascades.
