For immunofluorescence analysis, HeLa cells were fixed and permeabilized for detection of endogenous WWTR1 pS311 using Anti-Phospho-WWTR1 (Ser311) Recombinant Rabbit Multiclonal Antibody (ab277791, 1/100) and labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (1/2000). Nuclei (blue) were stained using ProLong Diamond Antifade Mountant with DAPI(Product # P36962) and cytoskeletal F-actin (red) staining using Rhodamine Phalloidin (1/300) Panel a-d) shows representative un-treated cells that were stained for detection and localization of Phospho-WWTR1 (Ser311) protein (green) with no signal compared to panel e-h) clearly demonstrating enhanced cytoplasmic localisation of WWTR1 pS311 in HeLa cells treated with Okadaic acid (1 M, 1h) and MG132 (40 M, 3h). The images were captured at 60X magnification.
