Function
May be involved in Ca(2+)-dependent exocytosis of secretory vesicles through Ca(2+) and phospholipid binding to the C2 domain or may serve as Ca(2+) sensors in the process of vesicular trafficking and exocytosis. -
Tissue specificity
Expressed in a variety of adult and fetal tissues. -
Sequence similarities
Belongs to the synaptotagmin family.
Contains 2 C2 domains. -
Domain
The second C2 domain/C2B is responsible for SYNCRIP binding. -
Cellular localization
Cytoplasmic vesicle > secretory vesicle > synaptic vesicle membrane. - Information by UniProt

All lanes : Anti-Synaptotagmin VII antibody (ab106618) at 1 µg/ml (Milk blocking - 3%)
Lane 1 : Brain (Mouse) Tissue Lysate
Lane 2 : Mouse Cortex Tissue Lysate
Lane 3 : Brain (Rat) Tissue Lysate
Lane 4 : Rat Cortex Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 45 kDa
Observed band size: 45,63,68 kDa why is the actual band size different from the predicted?
Additional bands at: 43 kDa, 76 kDa, 78 kDa. We are unsure as to the identity of these extra bands.
Exposure time: 12 minutes
The bands observed are representative of Synaptotagmin VII splice variants alpha, beta, and delta. While these differ to its predicted molecular weight of 45 kDa, this migration has been observed in the literature (PMID:17709608) This blot was produced using a 10% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% Milk before being incubated with ab106618 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
