SPHK1 was immunoprecipitated from HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate (1 mg for IP, 20% of IP loaded) with ab245354 at 10 μg/ml. Western blot was performed on the immunoprecipitate using ab245354 at 1 μg/ml.
Lane 1: ab245354 IP in HeLa whole cell lysate.
Lane 2: Control IgG in HeLa whole cell lysate.
Detection: Chemiluminescence at an exposure time of 30 seconds.
The more slowly migrating bands are believed to represent additional forms of SPHK1 owing to recognition by multiple antibodies each of which was directed toward distinctly different epitopes on SPHK1.
