Target

  • Function

Accessory subunit of the mitochondrial membrane respiratory chain NADH dehydrogenase (Complex I), that is believed not to be involved in catalysis. Complex I functions in the transfer of electrons from NADH to the respiratory chain. The immediate electron acceptor for the enzyme is believed to be ubiquinone.
  • Sequence similarities

Belongs to the complex I NDUFB8 subunit family.
  • Cellular localization

    Mitochondrion inner membrane.
  • Target information above from: UniProt accession O95169 The UniProt Consortium
The Universal Protein Resource (UniProt) in 2010
    Nucleic Acids Res. 38:D142-D148 (2010) .

      Western blot - Anti-NDUFB8 antibody (ab155903)
      All lanes : Anti-NDUFB8 antibody (ab155903) at 1 µg/ml

Lane 1 : Human heart tissue lysate - total protein (ab29431)
Lane 2 : Human Heart Mitochondrial Lysate
Lane 3 : Heart (Mouse) Tissue Lysate

Lysates/proteins at 10 µg per lane.

      Secondary
      All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution

      Developed using the ECL technique.

      Performed under reducing conditions.

      Predicted band size: 21 kDa
      Observed band size: 19 kDa
why is the actual band size different from the predicted?
Additional bands at: 37 kDa, 75 kDa. We are unsure as to the identity of these extra bands.

      Exposure time: 30 seconds

The band observed at 19 kDa could potentially be a cleaved form of NDUFB8 due to the presence of a 28 amino acid transit peptide. This blot was produced using a 10% Bis-tris gel under the MES buffer system. The gel was run at 200V for 35 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab155903 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
