All lanes : Anti-KSHV ORF45 antibody [2D4A5] (ab36618)
Lane 1 : Molecular weight marker
Lane 2 : Recombinant KSHV ORF45 expressed in E.coli
Predicted band size: 50 kDa
Observed band size: 78 kDa why is the actual band size different from the predicted?
Based on the analysis of the mRNA sequence, ORF45 is expected to encode a protein of 407 amino acids with a molecular mass of about 50 kDa. However, ORF45 was displayed as a diffuse band at the position of 78 kDa on a Western blot after SDS-PAGE. This observation implied that ORF45 might be highly post-translationally modified, resulting in its reduced mobility in denaturing SDS-PAGE. A survey of the primary amino acid sequence of ORF45 revealed multiple potential sites of phosphorylation, including a serine-rich region and other consensus sites for phosphorylation by the serine/threonine kinase, casein kinase II, and protein kinase C (Zhu & Yuan, 2003). -
