Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-HLA DRB1 + HLA-DPB1 antibody [JS76] (ab241560)
Immunohistochemical analysis of paraffin-embedded human tonsil tissue labeling HLA DRB1 + HLA-DPB1 with ab241560 at 0.798µg/ml, followed by Goat Anti-Mouse IgG H&L (HRP polymer) (ab214879) ready to use. Positive staining on human tonsil is observed. Counter stained with hematoxylin.
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20 mins.
The section was incubated with ab241560 for 30 mins at room temperature.
The immunostaining staining was performed on a Leica Biosystems BOND® RX instrument
Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Mouse IgG H&L (HRP polymer) (ab214879) ready to use.

Lane 1 : Ramos (Human Burkitt’s lymphoma B lymphocyte) lysate
Lane 2 : HeLa (human cervix adenocarcinoma epithelial cell) whole cell lysate

All lanes : Peroxidase-Conjugated Goat anti-Mouse IgG (H+L) at 1/100000 dilution
Predicted band size: 29 kDa
Observed band size: 26 kDa why is the actual band size different from the predicted?Blocking and dilution buffer: 5% NFDM/TBST.
Exposure time: 114 seconds.
Negative control: Hela (PMID: 20959457).
The molecular weight observed is consistent with what has been described in the literature (PMID: 20959457).
