Immunofluorescence analysis of ab272143. HeLa (Human epithelial cell line from cervix adenocarcinoma) cells were treated with primary antibody ab272143 at a 1:50 dilution for 1 hour at room temperature. FITC secondary antibody at 1:10,000 for 45 minutes at room temperature. Histone H3 [Trimethyl Lys4] localization is nuclear and chromosomal. Histone H3 K4me3 staining was expressed green, and the nuclei are counterstained blue with DAPI. Fixation was 0.5% PFA abd antigen retrieval was not required.

Chromatin from one million formaldehyde cross-linked HeLa (Human epithelial cell line from cervix adenocarcinoma) cells was used with 2 µg of ab272143 and 20µl of magnetic IgG beads per immunoprecipitation. A no antibody (No Ab) control was also used. Immunoprecipitated DNA was quantified using quantitative real-time PCR and SYBR green dye, then normalized to the non-precipitated input chromatin, which is equal to one.
