Three siRNA sequences targeting different sites of human FASTK cDNA and a scrambled control siRNA (si-NC) were transfected into U251 cells using Lipofectamine 2000. Total protein or total RNA was isolated at 48 h or 24 h post-transfection. FASTK protein levels were determined by western blot analysis using ab97544 at a dilution of 1/500 (A). The secondary antibody was a rabbit IgG-HRP and the quantitive data was obtained with the Quantity One analysis program. FASTK mRNA levels were assessed by qRT-PCR (B).
