Western blot protocol advice:
Since intracellular levels of pS294 ER are very low, Western blot without prior immunoprecipitation of total ER is not sensitive enough to detect pS294 ER. We recommend initial ER immunoprecipitation, following which it is possible to use this antibody for Western blot (see data image).

Exposure time: 3 minutesHarvested cells were lysed and immunoprecipitated using an ERα antibody; and the immunoprecipitated ERα (20μl each from un-transfected, ER alpha vector transfected, E2 and Din/E2 samples) was used to prepare separate western blots probed with either ab207602 anti-ERpS294 monoclonal or total ERα, all of which run at 67 kDa molecular weight. (PMID: 29137354).
