Observed band size: 75-100 kDa why is the actual band size different from the predicted?This data was developed using the same antibody clone in a different buffer formulation (ab199768).
Lanes 1 - 6: Merged signal (red and green). Green - ab199768 observed at 75-100 kDa. Red - loading control ab8245 (Mouse anti-GAPDH antibody [6C5]) observed at 37 kDa.
ab199768 was shown to react with DAG1 in Western blot. Membranes were blocked in fluorescent western blot (TBS-based) blocking solution before incubation with ab199768 and ab8245 (Mouse anti-GAPDH antibody [6C5]) overnight at 4 °C at 1 µg/ml and a 1 in 20000 dilution respectively. Blots were incubated with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 h at room temperature before imaging.

Extracellular matrix staining on mouse skeletal muscle (PMID: 29855340, PMID: 18691338) is observed. Counter stained with hematoxylin.
Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (HRP) ready to use.
Heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0).
