Immunohistochemical analysis of paraffin-embedded (Panel A) HEK-293T transfected with a 6X His tag® construct and (Panel B) HEK-293T transfected with empty plasmid, labeling 6X His tag® with ab252883 at 1/2000 dilution (0.498µg/ml) followed by ready to use Goat Anti-Mouse IgG H&L (HRP polymer) (ab214879). Positive staining on (Panel A) HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) transfected with a 6X His tag® construct, no staining on (Panel B) HEK-293T transfected with empty plasmid. The section was incubated with ab252883 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin. Secondary antibody only control: Secondary antibody is ready to use Goat Anti-Mouse IgG H&L (HRP polymer) (ab214879).
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

6X His tag® was immunoprecipitated from HEK-293T transfected with cas9 (S. Aureas) expression vector containing a myc-His-tag®, whole cell lysate with ab252883 at 1/30 dilution (33.167µg/ml) (2µg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab252883 at 1/1000 dilution (0.995µg/ml). VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.
Lane 1: HEK-293T transfected with cas9 (S. Aureas) expression vector containing a myc-His-tag®, whole cell lysate 10µg.
Lane 2: ab252883 IP in HEK-293T transfected with cas9 (S. Aureas) expression vector containing a myc-His-tag®, whole cell lysate.
Lane 3: Mouse monoclonal IgG2b instead of ab252883 in HEK-293T transfected with cas9 (S. Aureas) expression vector containing a myc-His-tag®, whole cell lysate.
