Alpha-L-Fucosidase (FUCA1) Assay Kit (Colorimetric) (ab273316) provides a simple, sensitive and high-throughput adaptable approach to detect physiological concentrations of this glycosidase in a variety of biological samples.
In this assay, Alpha-L-Fucosidase uses a synthetic chloro p-nitrophenol derivative (R-pNP) as an α-L-Fucosidase substrate and releases a chlorinated pNP derivative which can be measured kinetically under acidic conditions (OD 405 nm). The assay involves a one-step simple reaction, with minimal sample preparation and does not need a stop solution to complete the reaction.
The assay can detect as low as 20 µU of Alpha-L-Fucosidase activity in a variety of samples.

Alpha-L-fucosidase is responsible for hydrolyzing the alpha-1,6-linked fucose joined to the reducing-end N-acetylglucosamine of the carbohydrate moieties of glycoproteins. -
Involvement in disease
Defects in FUCA1 are the cause of fucosidosis (FUCA1D) [MIM:230000]. FUCA1D is an autosomal recessive lysosomal storage disease characterized by accumulation of fucose-containing glycolipids and glycoproteins in various tissues. Clinical signs include facial dysmorphism, dysostosis multiplex, moderate hepatomegaly, severe intellectual deficit, deafness, and according to age, angiokeratomas. -
