Rat L-Selectin ELISA Kit (ab263876) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of L-Selectin protein in cell culture supernatant, cit plasma, edta plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat L-Selectin with 0.77 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
L-Selectin, also referred to as CD62L and Leukocyte adhesion molecule 1, is a calcium-dependent lectin encoded by the gene SELL. L-Selectin binds to glycoproteins to mediate cell adhesion on neighboring cells. In peripheral lymph nodes, L-selectin mediates the adherence of lymphocytes to endothelial cells with high endothelial venules. L-Selectin is composed of several domains, one homologous to lectins, an epidermal growth factor domain, and two to repeat units found In C3/C4 binding proteins.

The L-Selectin standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of L-Selectin were measured in duplicates, interpolated from the L-Selectin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (EDTA) 100%, and plasma (citrate) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean L-Selectin concentration was determined to be 41.8 ng/mL in serum, 30 ng/mL in plasma (EDTA), and 25.1 ng/mL in plasma (citrate).

Undiluted samples are as follows: spleen cell supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean L-Selectin concentration was determined to be 44.1 ng/mL in spleen cell supernatant.
