Product overview
Rat IL-7 ELISA kit (ab277704) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Rat IL-7 protein in rat serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat IL-7 with 1.27 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
IL-7 is a cytokine important for B and T cell development. In humans, IL-7 and hepatocyte growth factor form a heterodimer that functions as a pre-pro-B cell growth-stimulating factor. IL-7 is found to be a cofactor for V(D)J rearrangement of the T cell receptor beta (TCRB) during early T cell development. It can be produced locally by intestinal epithelial and epithelial goblet cells and may serve as a regulatory factor for intestinal mucosal lymphocytes. Knockout studies in mice suggested that IL-7 plays an essential role in lymphoid cell survival. Rat IL-7 shares 89% and 59% sequence homology with mouse and human respectively.

Relevance
Interleukin 7 (IL-7) is a lymphoid cell growth factor that affects pre-B, pro-B, and early T cells. IL-7 was previously known as pre-B cell growth factor and lymphopoietin 1. IL-7 supports the growth of early B cells from long-term lymphoid bone marrow cultures. It is mitogenic to thymocytes and enhances the response of cells to other stimuli such as polyhydroxyalkanoate (PHA) and concanavalin A (ConA). IL-7 stimulates the proliferation of CD4+/CD8+ cells. The proliferative response of thymocytes to IL-7 is not affected by antibodies to the T cell growth factors such as IL2, IL4 and IL6, suggesting that IL-7 is capable of stimulating T cell proliferation through a pathway independent of the known T cell growth factors. Mature T cells respond to IL-7 and Con A, but not to IL-7 alone. In myeloid lineage cells, IL-7 upregulates the production of pro-inflammatory cytokines and stimulates the tumoricidal activity of monocytes/macrophages. IL-7 is expressed by adherent stromal cells from various tissues. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The IL-7 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of IL-7 were measured in duplicates, interpolated from the IL-7 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-7 concentration was determined to be 251.84 pg/mL in serum.
