Rat CXCL10 ELISA kit (ab270896) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CXCL10 protein in rat cell and tissue supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate rat CXCL10 with 87.45 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Rat C-X-C motif chemokine 10 (CXCL10) is a 10 kDa protein that is secreted by several cell types in response to interferon-gamma. These cell types include monocytes, endothelial cells and fibroblasts. Upon secretion, CXCL10 is cleaved into an 8.7kD biologically active protein to function in chemotaxis for T-cells, NK cells, monocytes/macrophages and dendritic cells. In addition, CXCL10 has antitumor activity through the inhibition of bone marrow colony formation and angiogenesis. CXCL10 elicits its effects by binding to the cell surface chemokine receptor 3 (CXCR3). Rat CXCL10 shares a 79% and 68% sequence identity with mouse and human, respectively.

The CXCL10 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of CXCL10 were measured in duplicates, interpolated from the CXCL10 standard curves and corrected for sample dilution. Undiluted samples are as follows: 5 ug/mL LPS stimulated spleen culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CXCL10 concentration was determined to be 6364.20 pg/mL in and stimulated spleen supernatant.
