Preparation of Substrate Solutions:
ELISA : 1M diethanolamin, 0.5mM MgCl2, 15mM pNPP, pH 9.8. Incubate for 15 minutes at 37°C. Stop the reaction with V/5 K2HP04 2M. Read at 405 nm.
Immunoblot : 34 µl of BCIP (5bromo-4chloroindolylphosphate) at 50mg/ml in DMF + 44 µl NBT (NitroBlue Tetrazolium) at 75 mg/ml in DMF 70% + 10ml substrate buffer (0.1M TRIS/HCI, 0.5M NaCl, 0.05M MgCl2, pH 9.5). -
