Plant selection genes in the pCambia vectors are driven by a double-enhancer version of the CaMV₃₅S promoter and terminated by the CaMV₃₅S polyA signal. Reporter genes feature a hexa-Histidine tag at the C-terminus to enable simple purification on immobilized metal affinity chromatography resins.

The GusPlus™ gene contains an intron from the castor bean catalase gene to prevent expression by bacteria and ensure detection of plant-expressed glucuronidase activity.

This vector also contains the rice glycine-rich protein signal peptide sequence (GRP) inserted between the NcoI and BglII sites.
