    pCambia1300 Plant Expression Vector
  • General notes

    Vector contains kanamycin resistance gene for bacterial selection, hygromycin B resistance gene for plant selection and no reporter gene to allow construction of user's own system.

    The pCambia1300 vector offers:
    High copy number in E.coli for high DNA yields
    pVS1 replicon for high stability in Agrobacterium
    Small size
    Restriction sites designed for modular plasmid modifications and small but adequate poly-linkers for introducing your DNA of interest
    Bacterial selection with kanamycin
    Plant selection with hygromycin B

    Plant selection genes in the pCambia vectors are driven by a double-enhancer version of the CaMV35S promoter and terminated by the CaMV35S polyA signal.

    This vector contains minimal heterologous sequences for plant transformation and selection of transformants, allowing insertion of desired genes for transformation into plants but require all promoter and terminator sequences for plant expression of newly cloned genes.

    The pUC18 polylinker within the lacZa fragment allows blue/white screening of clones in E. coli cloning work.

    In bacterial system:
    Kanamycin resistance (aphIII/nptIII/3’5’’-aminoglycoside phosphotransferase typeIII from
    Enterococcus faecalis pJH1) in bacteria: use 50 μg/mL in E.coli and A.tumefaciens.

    In plant system:

    Resistance Gene hph (From a Klebsiella species, via E.coli)
    Transformed plant selection antibiotic Hygromycin B
    Concentration in Arabidopsis 50 μg/mL in germination medium
    Concentration in Rice 50 μg/mL in 2N6 then in RGH6
    Concentration in Tobacco 50 μg/mL in RMOP then in MST
