Mouse uPA ELISA Kit (ab245727) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of uPA protein in cit plasma, edta plasma, hep plasma, serum, urine, and cell culture media. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse uPA with 31.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Urokinase type plasminogen activator (uPA) is secreted from cells as a precursor which is activated to the two chain form consisting of an A and B chain. The active high MW form is further processed by removal of an amino terminal fragment to an active low MW form (35 kDa). uPA is a serine protease that activates plasminogen toplasmin. High levels of uPA and plasminogen activator inhibitor type 1 (PAI 1) in breast cancer tissue extracts have been associated with rapid disease progression. The malignant phenotype of prostatic tumor cells correlates with the expression of both uPA and its cell membrane receptor (uPAR). -

The uPA standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of uPA were measured in duplicates, interpolated from the uPA standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, plasma (EDTA) 25%, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean uPA concentration was determined to be 7166 pg/mL in serum, 6222 pg/mL in plasma (citrate) and 7060 pg/mL in plasma (EDTA), and 4907 pg/mL in plasma (heparin).

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean uPA concentration was determined to be 272.6 ng/mL in urine.
