Abcam’s TNFSF4 Mouse ELISA Kit (ab193729) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse TNFSF4 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for mouse TNFSF4 coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures TNFSF4 present in the samples. The wells are washed and biotinylated anti-mouse TNFSF4 antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of TNFSF4 bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Involvement in disease
Genetic variations in TNFSF4 influence susceptibility to systemic lupus erythematosus (SLE) [MIM:152700]. SLE is a chronic, inflammatory and often febrile multisystemic disorder of connective tissue. It affects principally the skin, joints, kidneys and serosal membranes. It is thought to represent a failure of the regulatory mechanisms of the autoimmune system. Note=The upstream region of TNFSF4 contains a single risk haplotype for SLE, which is correlated with increased expression of both cell-surface TNFSF4 and TNFSF4 transcripts. Increased levels of TNFSF4 are thought to augment T cell-APC interaction and the functional consequences of T cell activation, thereby destabilizing peripheral tolerance. -
