Human NFAT2 ELISA kit (ab277724) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human NFAT2 protein in human cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human NFAT2 with 4 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Interleukin-2 Receptor is a plasma membrane receptor for interleukin-2. The receptor is involved in the regulation of immune tolerance by controlling regulatory T cells (TREGs) activity. TREGs suppress the activation and expansion of autoreactive T-cells. Interleukin-2 receptor is a non-covalent dimer of an alpha and a beta subunit. Interleukin-2 receptor exists in 3 different forms: a high affinity dimer, an intermediate affinity monomer (beta subunit), and a low affinity monomer (alpha subunit). The high and intermediate affinity forms also associate with a gamma subunit. This kit is designed to detect the soluble form of Interleukin-2 receptor subunit alpha (soluble IL-2 Receptor).

Relevance
The IL-2 Receptor, which is involved in T cell-mediated immune responses, alpha (IL-2RA) and beta (IL-2RB) chains, together with the common gamma chain (IL-2RG), constitute the high-affinity IL-2 receptor. Homodimeric alpha chains (IL-2RA) result in low-affinity receptor. Normally an integral-membrane protein, soluble IL-2RA has been isolated and determined to result from extracellular proteolyisis. Alternately-spliced IL-2RA mRNAs have been isolated, but the significance of each is presently unknown. Homodimeric beta (IL-2RB) chains produce a medium-affinity receptor. The intermediate affinity form consists of an alpha/beta subunit heterodimer, while the high affinity form consists of an alpha/beta/gamma subunit heterotrimer. Both the intermediate and high affinity forms of the receptor are involved in receptor-mediated endocytosis and transduction of mitogenic signals from interleukin 2. The interleukin 2 (IL-2) receptor gamma chain (IL-2RG), an important signalling component of many interleukin receptors (IL-2,IL-4,IL-7,IL-9, and IL-15), is thus referred to as the common gamma chain. Mutations in this X-chromosome-linked gene cause X-linked severe combined immunodeficiency (XSCID). -

The soluble IL-2 Receptor standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of soluble IL-2 Receptor were measured in duplicates, interpolated from the soluble IL-2 Receptor standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, plasma (heparin) 25% and spleen supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean soluble IL-2 Receptor concentration was determined to be 7,908 pg/mL in neat serum, 7,484 pg/mL in neat plasma (citrate), 5,442 pg/mL in neat plasma (EDTA), 7,172 pg/mL in neat plasma (heparin) and 837 pg/mL in neat spleen supernatant.
