Mouse MSP ELISA Kit (ab256390) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MSP protein in cit plasma, edta plasma, hep plasma, serum, urine, tissue lysate, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse MSP with 129.22 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Macrophage stimulating protein (MSP) is a serum protein also known as hepatocyte growth factor-like that is encoded by the MST1 gene. Secreted by the liver as the main source, MSP has the ability to exert biological effects such as proliferation, migration, differentiation, and phagocytosis through binding to the receptor tyrosine kinase. The standard protein in this kit is a fragment of the full length protein that contains the alpha chain.

Relevance
Macrophage Stimulating Protein contains four kringle domains and a serine protease domain, similar to that found in hepatic growth factor. Despite the presence of the serine protease domain, the encoded protein may not have any proteolytic activity. The receptor for this protein is RON tyrosine kinase, which upon activation stimulates ciliary motility of ciliated epithelial lung cells. This protein is secreted and cleaved to form an alpha chain and a beta chain bridged by disulfide bonds. -

The MSP standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of MSP were measured in duplicates, interpolated from the MSP standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:400, plasma (citrate) 1:200, plasma (EDTA) 1:400 and plasma (heparin) 1:400. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MSP concentration was determined to be 6,320 ng/mL in serum, 1,580 pg/mL in plasma (citrate), 4,435 pg/mL in plasma (EDTA), and 4,262 pg/mL in plasma (heparin).

The concentrations of MSP were measured in duplicates, interpolated from the MSP standard curves and corrected for sample dilution. Undiluted samples are as follows: urine 100% and liver supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MSP concentration was determined to be 1,895 pg/mL in urine and 37,547 pg/mL in liver supernatant.
